My point, manually shifting vs auto is to rule out a possible electrical problem.
(first I always recommend to drive in 3 unless you are driving 55mph+)
Now to the op's issue, if you start driving in 1, then shift to 2 @15mph and it kicks in, then shift to 3 @25 and it kicks in, then there is a good chance that there is a fluid flow issue through the shift solenoids. Drop the pan, clean it out, change the filter, and install fresh dexVI.
